The need for quantitative and qualitative analysis of phytohormones in a wide variety of plant materials is growing everyday by more biologists with multiple interests, both fundamental and applied.

Phytohormones that can be extracted, purified and assayed are auxins, cytokinins, polyamines.

Auxins

The analysis of the different auxins, simultaneously, is carried out on samples of 500 mg of fresh matter or frozen with liquid nitrogen, and three repetitions are advised. The assays are carried out in UPLC using the natural fluorescence of auxins.

The various quantifiable auxins are indole-3-acetic acid (AIA), AIA aspartate, AIA alanine, AIA phenylalanine, glutamic AIA, indole-3-butyric acid (AIB) and  AIB alanine.

Cytokinins (liquid extract)

The analysis of the different cytokinins, simultaneously, is carried out on samples of 2 g of fresh material or liquid nitrogen frozen, and three replications are recommended. The assays are carried out in UPLC coupled to a diode arrays detector.

The various quantifiable cytokinins are zeatin, zeatin riboside, 2-isopentenyladenine (2-iP), 2-isopentenyladenosine (2-iPA), benzylaminopurine (BAP) and BAP riboside.
